Isolation and characterization of a novel Escherichia coli bacteriophage vB_EcoM-785 and functional analysis of its lysin Lys vB and holin Hol 785

Escherichia coli, rod-shaped and gram-negative bacteria belonging to the Enterobacteriaceae family, is a significant concern in public health and food safety, which is capable of toxin secretion, aggregative colonization, multiplying in the gastrointestinal tract, and damaging different environments. The common treatment is antibiotic therapy; however, the misuse of antibiotics has led to the continuous emergence of drug-resistant strains, which pose a severe threat to public health by limiting clinical treatment and prophylactic options. With the disadvantages of traditional antimicrobial agents, novel strategies are under development to fight multidrug-resistant strains. Among them, bacteriophages have been successfully exploited to treat some infections of pathogens. Lytic phages are preferred candidates for killing bacteria and they have indeed achieved great success in fighting against multidrug-resistant bacteria, with no negative impact on the normal flora and organismal cells in the animal. Therefore, the purpose of this study was to isolate and screen a strain of Escherichia coli phage vB_EcoM-785 with good cleavage activity and stability. After whole genome sequencing and analysis, the target proteins Lys vB and Hol 785 of phage vB_EcoM-785 were cloned, expressed and purified. The antibacterial activity and their practical application of phage vB_EcoM-785 and its derived enzymes were explored.
